Exosomal miR-186 derived from BMSCs promote osteogenesis through hippo signaling pathway in postmenopausal osteoporosis

Abstract Background Postmenopausal osteoporosis (PMO) that results from estrogen withdrawal is the most common primary among older women. However, little known about mechanism of PMO, and effective treatment PMO limited. Methods We used real-time polymerase chain reaction (qPCR), Western blotting, RNA pull down to investigate relationship between miR-186 MOB Kinase Activator 1A (Mob1). Also, we investigated effect exosome in osteogenesis using alkaline phosphatase (ALP) staining. And hematoxylin eosin (HE) staining was verify model. Results Exosomal plays an important role bone formation. The miRNA-seq q-PCR showed upregulated a + Exo group. RNA-pull luciferase reporter assays verified interactions Mob1. also Hippo signaling pathway osteogenesis. Conclusions concluded exosomes derived human marrow mesenchymal stem cells (hBMSCs) can transfer promote ovariectomy (OVX) rats through pathway.
